HIV Drugs May Be Tied to Early Aging (HealthDay)

SUNDAY, June 26 (HealthDay News) -- An older class of antiretroviral drugs used to treat HIV can cause premature aging, a new study suggests.

Researchers examined muscle cells from HIV patients and found that zidovudine (AZT) and other antiviral drugs known as nucleoside analogue reverse-transcriptase inhibitors (NRTIs) damage DNA in mitochondria, the energy factories in cells.

The study is published June 26 in the journal Nature Genetics.

The finding may help explain why some HIV patients treated with antiviral drugs show advanced signs of frailty and age-related diseases such as dementia and cardiovascular disease at an early age.

"HIV clinics were seeing patients who had otherwise been successfully treated but who showed signs of being much older than their years. This was a real mystery," Professor Patrick Chinnery, a senior fellow in clinical science at the Institute of Genetic Medicine at Newcastle University in England, said in a Wellcome Trust news release.

"The DNA in our mitochondria gets copied throughout our lifetimes and, as we age, naturally accumulates errors," he explained.

"We believe that these HIV drugs accelerate the rate at which these errors build up. So over the space of, say, 10 years, a person's mitochondrial DNA may have accumulated the same amount of errors as a person who has naturally aged 20 or 30 years. What is surprising, though, is that patients who came off the medication many years ago may still be vulnerable to these changes."

Because they're relatively cheap, NRTIs are important for people in Africa and low-income countries, said study co-author and HIV specialist Dr. Brendan Payne of the Royal Victoria Infirmary in Newcastle.

"These drugs may not be perfect, but we must remember that when they were introduced they gave people an extra ten or twenty years when they would otherwise have died," Payne said in the news release. "In Africa, where the HIV epidemic has hit hardest and where more expensive medications are not an option, they are an absolute necessity."

Usually a reference to a peculiarly American, public high school, curricular program or ?track,? offering a variety of ?self-improvement? courses and rudimentary courses in English, mathematics, science and social studies. The term general education was first used and defined in a landmark 1945 report by the Harvard Committee, General Education in a Free Society. The report defined a core curriculum that all students, whether pursuing vocational or academic education, should, ?as citizens and heirs of a joint culture,? study. It consisted of three years each of English, mathematics and science and two years of social studies. The term lost its meaning as many public high schools adopted ability-grouping programs that segregated students into so-called tracks, with the highest academic achievers in the college-bound or honors track. In 1994, Congress tried to end general education by passing the SCHOOL-TO-WORK-OPPORTUNITIES ACT, a federal law requiring states to establish education programs in all public schools to prepare students either for continuing education or provide them with the skills necessary for gaining entry into the job market after graduating from high school. In effect, the law ended all federal funding for general education, which provides neither of these outcomes, and it forced schools to begin developing effective new curricula for work-related vocational education to guarantee each student either a high school diploma (or equivalent), a nationally recognized skill certificate or an associate degree that will lead to a first job or further education. Funded with more than $1.5 billion in federal monies, the School-To-Work Opportunities Act provided financial and legislative impetus to the specialized ?VO-TECH? and COOPERATIVE VOCATIONAL EDUCATION programs already functioning in about 500 forward-looking public school districts.

Floodwater seeps into Nebraska nuke plant building (AP)

OMAHA ? Missouri River floodwater seeped into the turbine building at a nuclear power plant near Omaha on Monday, but plant officials said the seepage was expected and posed no safety risk because the building contains no nuclear material.

An 8-foot-tall, water-filled temporary berm protecting the plant collapsed early Sunday. Vendor workers were at the plant Monday to determine whether the 2,000 foot berm can be repaired.

Omaha Public Power District spokesman Jeff Hanson said pumps were handling the problem at the Fort Calhoun Nuclear Station and that "everything is secure and safe." The plant, about 20 miles north of Omaha, has been closed for refueling since April. Hanson said the berm's collapse didn't affect the shutdown or the spent fuel pool cooling.

Nuclear Regulatory Commission spokesman Victor Dricks described the situation as stable. NRC Chairman Gregory Jaczko plans to inspect the Fort Calhoun plant on Monday as part of a pre-arranged visit to Nebraska.

Hanson said OPPD fired up generators and cut the power supply after water surrounded the main electrical transformers on Sunday. The generators powered the plant until an off-site power supply was connected later in the day.

Officials said the berm wasn't critical to protecting the plant, which sits across the river from Iowa.

"There are other structures and systems in place that can ensure they will continue operating safely," Jaczko said Sunday.

The river was not expected to rise higher than the level the plant is designed to handle.

Jackzo inspected the Cooper Nuclear Station, which sits on the Missouri River about 75 miles south of Omaha, on Sunday. He asked plant officials and the NRC's local inspectors questions about the plant and this year's flooding.

The plant, which is owned by Nebraska Public Power District, remains dry because it sits at an elevation above the river and continues to operate at full capacity. The base of Cooper and its storage area for used nuclear fuel is 903 feet above sea level. The river was 900.2 feet above sea level early Monday.

Both nuclear plants issued flooding alerts earlier this month, although they were routine as the river's rise has been expected.

Flooding remains a concern all along the Missouri because of massive amounts of water the U.S. Army Corps of Engineers has released from upstream reservoirs. The river is expected to rise as much as 5 to 7 feet above flood stage in much of Nebraska and Iowa and as much as 10 feet over flood stage in parts of Missouri.

The corps expects the river to remain high at least into August because of heavy spring rains in the upper Plains and substantial Rocky Mountain snowpack melting into the river basin.

Iranian oil minister says OPEC strains solvable (AP)

VIENNA ? Iran's oil minister on Monday acknowledged strains within OPEC after its last meeting exposed deep rifts between his country and rival Saudi Arabia, but said the organization can solve them internally.

The comments by Mohammad Aliabadi and other OPEC officials appeared to be an attempt to patch up the image of the 12-nation oil producing organization after it ended an abortive June 6 meeting split into two camps ? one led by Iran, the other by the Saudis. Iran, which holds the OPEC rotating presidency, wanted to keep output steady and supplies tight. The Saudis sought a decision to increase levels ? and they and their Gulf allies have since moved to boost production by themselves, shattering the image of OPEC unity.

The unprecedented rift appeared in part to be political between the two Middle East rivals. But both Ailabadi and other OPEC officials sought Monday to play down any differences at a briefing at the end of annual OPEC and EU talks meant to find common ground on production and prices.

OPEC member countries may "have differences," Aliabadi told reporters. "OPEC, however, ... is a family and solves its problems within the family."

Sunni-ruled Saudi Arabia has long vied with Shiite-rule Iran for regional dominance and the two have often been at loggerheads over pricing. In the past, however, OPEC, usually fell in behind Saudi Arabia, which produces the lion's share of OPEC output.

Tensions have recently been exacerbated by an invitation from Bahrain's Sunni rulers for a Saudi-led Gulf force to help suppress unrest by Bahrain's Shiite population, infuriating Iran.

Sunni-dominated Arab countries, in turn, fear gains by Bahrain's Shiites could allow Iran to expand its influence.

While both countries have been careful in the past to keep their rivalries out of public view, the Mideast unrest has sharpened tensions ? with OPEC caught in the middle.

In comments first reported by the Wall Street Journal, Saudi Prince Turki al-Faisal suggested this month that his countries could use oil policy to hobble Iran. "Iran is very vulnerable in the oil sector, and it is there that more could be done to squeeze the current government," Turki, Riyadh's former ambassador to Washington and London, was quoted as telling U.S. and British servicemen at an air base outside London.

Saudi Arabia could easily make up for any reduction of Iranian oil exports, due to sanctions or other measures enacted to pressure Iran over its nuclear program, Turki was quoted as saying, adding that Riyadh has enough spare production capacity to "instantly replace all of Iran's oil production."

Aliabadi said Monday he could not comment on Turki's reported comments. But OPEC Secretary-General Abdullah Al-Badri said the OPEC rift that doomed the June 6 meeting reflected a difference of views over production and "has nothing to do with political" issues.

Both officials were critical of the decision by industrialized countries to start releasing 60 million barrels of oil from emergency supplies after OPEC failed to agree to up output levels.

"The stocks are supposed to be for emergencies only and not for commercial activities," Al-Badri said, suggesting the move was an attempt to influence markets at a time when stocks were plentiful and prices reasonable.

Aliabadi said that OPEC is capable of meeting market requirements. "We believe that the IEA ... should not intervene commercially," he said, referring to the Paris-based International Energy Agency, which coordinated the oil release. In announcing the move Thursday, the agency said it was pre-emptive to prevent a sudden spike in prices.

Even before the oil release, oil prices tumbled from highs of more than $100 a barrel, with the Saudi-led production increases easing concerns about supply. They fell further after the IEA announced that it will put the extra barrels on the market over a 30-day period, with half coming from the U.S. Strategic Petroleum Reserve.

'The Muppets' Movie: All The Stills From New Jason Segel Disney Movie (PHOTOS)

While this summer has had no shortage of star power and big money films thus far, perhaps the movie creating the biggest buzz this season is a movie that won't be coming out until Thanksgiving.

Then again, it's been a long wait to get even this close.

A decade since their last big screen adventure, Kermit and his Muppet pals are getting a big screen reboot that hearkens back to their great showbiz musical tradition. Written by and starring Jason Segel (and co-starring Amy Adams), the film has been on a viral marketing tear the past few months, first releasing hilarious spoofs on current hit films (the romantic comedy, The Hangover, and Green Lantern), and then, unveiling its own teaser trailer.

Each one has been funnier than the next, but also true to the Muppet legacy of creator Jim Henson. And that's exactly what Segel has been going for.

"To me, I just really wanted to pay homage to those movies that meant so much to me growing up," he recently told a group of reporters. "I wanted the experience, like we talked about, where parents... Our generation of kids, we'll remember those great movies, those great moments. And then also being able to have our age introduce our kids to the Muppets. So yeah, that was my goal."

Biden offers voice of empathy on the economy (AP)

COLUMBUS, Ohio ? The longest walk parents can make, Vice President Joe Biden often says, is up a flight of stairs to tell their children they've lost their job.

"Millions have been stripped of their dignity," Biden told an Ohio audience last year. "It's time to restore their dignity."

Biden, who spoke frequently of his blue-collar roots in Scranton, Pa., during the 2008 presidential campaign, is reprising his role as one of the Obama administration's top surrogates on the economy and an empathetic voice in states throughout the industrial Midwest hard hit by the economic recession.

The former Delaware senator is expected to play a similar role in the 2012 campaign, focusing on states like Ohio, Michigan and Pennsylvania, which President Barack Obama carried in 2008 but which elected Republican governors in 2010. A large swath of the Midwest, including Iowa, Indiana and Wisconsin ? all won by Obama in 2008 ? are considered prime targets for Republicans next year.

"He obviously has deep, deep roots in the industrial Midwest running from Pennsylvania right across and he'll be very valuable there," Obama strategist David Axelrod told reporters in Chicago earlier this week. "But he's somebody we would send and will send anywhere in this country."

Biden, who was speaking Saturday to the Ohio Democratic Party's annual State Dinner, has assailed moves by Republican governors in Wisconsin and Ohio to strip away collective bargaining rights from most public workers and efforts by Republican lawmakers in Congress to turn Medicare into a program in which the government subsidizes beneficiaries who would seek coverage from private insurance companies.

The vice president has defended Obama's handling of the economy, pointing to tough decisions to seek an economic stimulus package and rescue U.S. automakers. But his pitch often turns personal, drawing on his father's decision to move the family to Delaware in the 1950s in search of a job.

"There's still a long way to go. There are still millions of women and men who are like the family I was raised in," Biden told Democrats in New Hampshire last month. "When a recession hit, we knew someone sitting around my dad's kitchen table ... was going to lose their job."

Former Ohio Gov. Ted Strickland, a Democrat who also rose from modest circumstances, said Biden "has the ability to express what a lot of people feel when it comes to their anxieties over the economy and job loss, their kids. I think he does a really good job of identifying with those concerns and expressing them."

Largely under the radar, Biden has maintained a busy travel schedule, appearing in more than 150 political events in 2009 and 2010, including 20 events in Ohio alone and 14 in Pennsylvania. In 2011, he had appeared at more than a dozen political events before Saturday's dinner in Ohio and is headlining fundraisers on Monday in Atlanta and Nashville, Tenn.

In July, Biden is scheduled to speak to the International Brotherhood of Teamsters in Las Vegas, the National Education Association annual convention in Chicago and the National Association of Police Organizations in Dallas. Next fall, he is addressing the Florida Democratic Party's state convention.

Don't be afraid to hire a financial adviser to set up and balance your retirement portfolio for you, either. Especially in the wake of the Madoff scandal, however, you'll definitely need to vet any financial pros you consider. One resource you can check to make sure a broker is legit is the Financial Industry Regulatory Authority (FINRA)'s database, and there are a few other resources mentioned by Forbes for avoiding that sort of major financial disaster.

Review and Update Your Plan Regularly

Although the tools above can give you some guidance and a plan of action, a comfortable retirement really is a goal that's hard to plan for decades in advance. Just as we now have longer lifespans that make retiring at age 65 less feasible, the market and other economic factors will continue to change. EU to pressure Greece amid bank rollover talks (Reuters)

BRUSSELS/ATHENS (Reuters) ? European Union leaders will pile pressure on Greece at a summit on Thursday to adopt deeply unpopular austerity measures in return for fresh funds to avert a bankruptcy that could shake the global economy.

Euro zone governments are meanwhile arm-twisting banks and insurers to maintain their exposure to Greek sovereign debt when their bonds mature, despite the heightened risk of default, as part of a planned second financial rescue for Athens.

Combining brow-beating and moral support, leaders will tell Greek Prime Minister George Papandreou they will release the next 12 billion euros ($17.2 billion) in emergency aid on July 3, to prevent Athens running out of money in mid-July, provided the Greek parliament adopts key economic reforms next week.

While he has expressed confidence over that vote in public, Slovak Prime Minister Iveta Radicova said Papandreou had voiced doubts in a private telephone call.

"Prime Minister Papandreou has serious doubts about whether the necessary steps will pass in parliament," Radicova told the Slovak parliament's European affairs committee.

Inspectors from the European Commission, European Central Bank and International Monetary Fund met new Greek Finance Minister Evangelos Venizelos in an effort to iron out differences on the bailout program, which he has said he wants to amend to appease an angry Greek public.

"Venizelos will not go to Brussels. He will continue the negotiations with the troika," a lawmaker who took part in a parliamentary committee with the minister told Reuters. "There is a gap of 3.8 billion euros out of the total package of 28 billion euros (in the mid-term fiscal plan) which should be discussed with the troika."

The Greek crisis is set to dominate the fourth EU summit this year as the 27 leaders grope for a solution to the debt woes that have forced Greece, Portugal and Ireland to seek bailouts, diplomats said.

German Chancellor Angela Merkel underlined that no formal decisions on Greece will be taken at the meeting, but the gathering will be monitored intensely by financial markets for any message it sends on whether the EU plan can work.

Investors are skeptical: Five-year credit default swaps on Greek government debt rose 138 basis points to 2,025 bps, according to data monitor Markit, implying a more than 80 percent probability of default over that period.

U.S. Federal Reserve Chairman Ben Bernanke stressed on Wednesday that much more than the future of Greece was at stake.

"If there were a failure to resolve that situation, it would pose threats to the European financial system, the global financial system, and to European political unity, I would conjecture, as well," he said.

The summit agenda also involves appointing Mario Draghi as the next head of the European Central Bank, agreeing to increase the size of the euro zone's current bailout fund, and completing the creation of a permanent crisis fund from June 2013.

STRESS THE POSITIVE

In an attempt to offer positive incentives to Greeks, who have staged mass protests against pay and pension cuts and are resisting privatizations demanded by the EU and IMF, European officials stressed their support for Athens' recovery.

"My message to the Greek people is that, if the government acts, Europe will deliver. If Greece can demonstrate that it is genuinely committed to the reform package ... we will accompany Greece on its journey back to growth," European Commission President Jose Manuel Barroso said on Wednesday.

Papandreou's reshuffled government won a confidence vote in parliament early on Wednesday, clearing one of several hurdles on the path to avoiding a default.

On June 28, parliament will vote on a package of spending cuts, tax increases and privatization measures that Athens has agreed with the EU and IMF. Euro zone finance ministers will review progress on July 3 with a view to releasing the tranche.

Despite EU and IMF calls for Greek political leaders to unite behind the program, all opposition politicians voted against the government in the confidence vote, and 20,000 protesters chanted insults outside parliament.

Even if Greece manages to persuade the EU and IMF that it is fully committed to making the budget adjustments demanded, this will buy the government only a few months' respite and most economists expect it will have to default eventually.

Greece accepted a package of 110 billion euros of EU/IMF loans in May 2010 and now needs a second bailout of a similar size to meet its financial obligations until the end of 2014, when it hopes to return to capital markets for funding.

Euro zone member states, led by Germany, insist any second aid package to include the involvement of the private sector.

At meetings on Wednesday, banks and insurers in Germany, France, Spain and Belgium and possibly other countries were asked by their national central banks to roll over their holdings of Greek debt when the bonds mature, banking and government sources said.

Belgium's De Tijd newspaper said Belgian private bondholders were asked to buy new five-year bonds at the euro zone rescue fund's lending rate upon maturity.

There was no official confirmation of the terms, but a spokeswoman for Franco-Belgian banking group Dexia said it is prepared to roll over its 5.4 billion euro exposure to Greek debt, the biggest among Belgian banks.

Credit ratings agencies have said that even a voluntary rollover would be classified as a default, which would have a profound impact on European and global financial markets.

The medium-term economic reform program agreed by Athens envisages raising 50 billion euros by selling off state firms and includes 6.5 billion in spending cuts and tax rises in 2011.

Even if Greece achieves its targets -- and it has already missed many objectives set by its international lenders -- it will still not be in a position to manage its debts, which already account for 150 percent of gross domestic product.

Breakthrough in the search for new treatments for multiple sclerosis

Scientists at the University of Nottingham have discovered a molecular mechanism which could bring about the development of new treatments for Multiple Sclerosis (MS) ? a chronic inflammatory disease of the central nervous system.

Dr Bruno Gran, a Clinical Associate Professor in the Division of Clinical Neurology in the School of Clinical Sciences, working in collaboration with Professor Paul Moynagh from the National University of Ireland, Maynooth, has discovered a synthetic chemical compound which inhibits the pro-inflammatory signals produced by the immune system in MS. What makes this chemical unique is that at the same time, it stimulates the body to produce interferon-beta, an anti-inflammatory molecule, that is commonly given to patients as an injected drug to treat MS.

Together, these effects cause significant reduction in the severity of an animal model of MS. the researchers have also discovered that cells of the immune system obtained from the blood of people with MS are more sensitive to the effects of this drug than those obtained from people who do not have MS.

Dr Gran said: ?Under laboratory conditions we have found a way of encouraging the body to produce its own Interferon-beta. when other experimental substances have been tested in the laboratory to achieve this effect, they usually cause the immune system to produce a mixture of anti-inflammatory as well as pro-inflammatory molecules, typically reducing the overall efficacy. In the case of the compound tested in this study (a synthetic cannabinoid known as R(+)WIN55,212-2), the predominantly anti-inflammatory effects appear promising for further pre-clinical, and hopefully clinical, testing.

With no available cure MS is the focus of intense study for the hundreds of scientists across the world who are working on new treatments for this disabling disease. MS is more common in temperate climates. with around 100,000 people suffering from MS in the UK the country has one of the highest rates of the disease in the world.

Until 20 years ago there was little progress in the search for treatments.

After their first approval in 1993 Beta Interferons still represent one of the first line treatments for relapsing-remitting multiple sclerosis. these drugs are not a cure but they can reduce the number and severity of relapses. Despite this, more effective, well tolerated therapeutic strategies are needed.

Dr Gran?s research continues a line of investigation which his laboratory has carried out for a number of years on the role of endogenous type I interferons in regulating multiple sclerosis inflammation in the central nervous system.

The cause of MS is still something of a mystery. Numerous factors are thought to contribute, including genetic susceptibility and environmental factors. the latter are thought to include certain viral infections and low levels of vitamin D, linked to poor sun exposure.

These latest findings highlight a new selective mechanism that may be open to exploitation in the development of new therapeutics for the treatment of MS.
